Reconstitution Basics for Lyophilized Peptides
Lyophilized (freeze-dried) peptides arrive as a stable powder. Reconstitution is the process of dissolving that powder into solution for use in a research model.
The standard diluent is bacteriostatic water, which contains 0.9% benzyl alcohol to inhibit microbial growth across repeated draws. Sterile or sodium-chloride water can be used for single-use work but lacks the preservative.
Choosing a reconstitution volume is a concentration decision, not a fixed rule. A common approach is to add a volume that makes the math clean — for example, 2 mL of bacteriostatic water into a 10 mg vial yields 5 mg/mL.
Add diluent slowly down the side of the vial rather than directly onto the powder, and let it dissolve without vigorous shaking. Once reconstituted, store refrigerated and protected from light.
